基于SSR标记对5个新疆陆地棉品种的指纹图谱构建及遗传多样性分析

Fingerprint map construction and genetic diversity analysis of 5 Xinjiang upland cotton varieties based on SSR markers

    目的 利用简单重复序列(SSR)分子标记技术,构建审定品种源棉8号及4个自育陆地棉品种的指纹图谱,并进行遗传多样性分析。
    方法 利用筛选出的64对核心引物,对源棉8号以及新疆4份自育陆地棉品种材料进行SSR分子标记检测,并构建指纹图谱及分析遗传多样性。
    结果 64对多态性SSR引物在5份棉花品(系)种中共扩增出88条多态性条带,观测等位基因数(Na)为128个,各位点Shannon’s多态性信息指数(I)范围为0.50040.6730,Nei’s 基因多样性指数(H)平均为0.4375。遗传多样性分析结果显示,源棉8号与源棉1278的遗传相似性系数最高,与源棉1280的遗传相似性系数最低,5份种质材料间的遗传距离范围为 0.37300.8133,总体表现出较低的遗传多样性。基于SSR标记构建的5个棉花品种的指纹图谱信息并生成专属二维码,可有效区分不同棉花品种。
    结论 以源棉8号为代表的5份新疆陆地棉种质资源间遗传多样性较低,本研究构建的5份棉花种质资源的SSR指纹图谱及二维码,可用于品种鉴定。

     

    Abstract:
    Objective This study aimed to construct DNA fingerprints of the certified variety Yuanmian 8 and four self-bred Gossypium hirsutum varieties using simple sequence repeat (SSR) molecular markers, and to analyze their genetic diversity.
    Methods Sixty-four pairs of screened core primers were used for SSR molecular marker detection on Yuanmian 8 and four self-bred upland cotton varieties from Xinjiang. DNA fingerprints were constructed and genetic diversity was analyzed.
    Results A total of 88 polymorphic bands were amplified by 64 polymorphic SSR primers across the five cotton varieties (lines). The observed number of alleles (Na) was 128. The Shannon’s information index (I) per locus ranged from 0.5004 to 0.6730, and the average Nei’s gene diversity index (H) was 0.4375. Genetic diversity analysis showed that the genetic similarity coefficient between Yuanmian 8 and Yuanmian 1278 was the highest, while that between Yuanmian 8 and Yuanmian 1280 was the lowest. The genetic distance among the five germplasm materials ranged from 0.3730 to 0.8133, indicating a low level of overall genetic diversity. Exclusive QR codes were generated based on the fingerprint information of the five cotton varieties constructed by SSR markers, which could effectively distinguish different cotton varieties.
    Conclusion The five Xinjiang upland cotton germplasm resources represented by Yuanmian 8 exhibit low genetic diversity. The SSR fingerprints and QR codes constructed in this study can be used for variety identification of cotton.
