不同硒浓度下人工栽培富硒羊肚菌的氨基酸特性及硒形态鉴定

Amino acid characteristics and selenium speciation of artificially cultivated selenium-enrich Morchella sextelatam under different selenium concentrations

    目的 通过对不同硒浓度下栽培的羊肚菌(Morchella sextelatam)进行氨基酸含量测定及硒形态鉴定分析,明确主要硒形态及不同浓度硒处理对羊肚菌中氨基酸的影响。
    方法 本研究在不同浓度外源硒处理条件下进行人工栽培羊肚菌,测定其子实体中多种氨基酸含量。通过相关性分析、主成分分析等,对羊肚菌各种氨基酸之间的相关性以及硒浓度对羊肚菌中氨基酸综合水平的影响进行分析,并进行硒形态鉴定。
    结果 研究结果表明,硒浓度处理为0.0~40.0 mg/L条件下,羊肚菌中16种氨基酸总含量在20.86%~22.53%之间;在硒浓度处理为40.0 mg/L时,必需氨基酸在总氨基酸中的占比最高,为38.45%。在不同硒浓度处理下,羊肚菌中鲜、甜味氨基酸总含量比例均高于苦味氨基酸含量比例,以鲜、甜味为主。羊肚菌各氨基酸之间绝大多数呈正相关,部分氨基酸之间呈显著相关(P<0.05),部分氨基酸之间呈极显著相关(P<0.01)。随着硒浓度处理的增加,羊肚菌中各种氨基酸含量的综合水平降低,添加外源硒会影响氨基酸的含量。羊肚菌子实体中硒的含量随着硒浓度的增加而增加,在硒浓度处理为20.0和40.0 mg/L条件下,其子实体主要硒形态为硒代甲硫氨酸,均未检出硒代胱氨酸、亚硒酸盐、硒酸盐;在硒浓度处理为20.0 mg/L时,硒代甲硫氨酸含量为1.980 mg/kg,占总硒含量的17.89%;在硒浓度处理为40.0 mg/L时,硒代甲硫氨酸含量为8.000 mg/kg,占总硒含量的24.94%。
    结论 综上所述,在硒浓度为20.0和40.0 mg/L处理下,羊肚菌中必需氨基酸含量均较高,各种氨基酸含量综合水平相对较低;硒代甲硫氨酸为主要硒形态,其含量分别为1.980、8.000 mg/kg。本研究为后续优化富硒羊肚菌栽培和开发富硒产品提供了技术支持。

     

    Abstract:
    Objective By further measuring the amino acid content and identifying the selenium forms of Morchella sextelatam under different selenium concentrations, the suitable selenium concentration for artificial cultivation of Morchella sextelatam was ultimately selected, providing methodological guidance for the further development of selenium rich Morchella sextelatam products.
    Methods This study artificially cultivated Morchella sextelatam under different concentrations of exogenous selenium, measured the content of various amino acids in their fruiting bodies, and evaluated the correlation between various amino acids in Morchella sextelatam and the effect of selenium concentration on the comprehensive level of amino acids in Morchella sextelatam through correlation analysis and principal component analysis. Selenium forms were also identified.
    Results The results showed that when the concentration of selenium was 0.0~40.0 mg/L, the total amino acid content of Morchella sextelatam was between 20.86% and 22.53%, among which the maximum was at 0.0 mg/L selenium, and the minimum was at 40.0 mg/L selenium. When selenium concentration was 40.0 mg/L, the proportion of essential amino acids in total amino acids showed the highest, which was 38.45%. Through the analysis of flavor amino acid composition, the total proportion of fresh sweet amino acids in the four groups of Morchella sextelatam was higher than that of bitter amino acids, with fresh sweet as the main flavor. Through correlation analysis, it was found that the majority of amino acids in Morchella are positively correlated, with some amino acids showing significant correlation (P<0.05) and some amino acids showing extremely significant correlation (P<0.01). According to the comprehensive evaluation of principal component analysis, with the increase of selenium concentration, the comprehensive level of various amino acids in Morchella sextelatam decreased, and the addition of exogenous selenium would affect the content of amino acids. According to the detection of Se element in fruit bodies, it was found that the content of Se needed by human body increased along with the increase of selenium concentration. SeCys2, Se(Ⅳ) and Se(Ⅵ) were not detected in the fruiting bodies of Morchella sextelatam at selenium concentrations of 20.0 and 40.0 mg/L. When the selenium concentration was 20.0 mg/L, the SeMet content was 1.98 mg/kg, accounting for 17.89% of the total selenium content; when the selenium concentration was 40.0 mg/L, the SeMet content was 8.00 mg/kg, accounting for 24.94% of the total selenium content. Based on the results of amino acid contents and selenium forms, 20.0 mg/L of selenium concentration was most suitable for artificial cultivation of selenium-enrich morel.
    Conclusion In summary, selenium concentrations of 20.0 and 40.0 mg/L can both be used as suitable selenium concentrations for selenium rich artificial cultivation of Morchella sextelatam. The essential amino acid content in the treated Morchella sextelatam is relatively high, and the overall level of various amino acid contents is relatively low; SeMet is the main form of selenium, with contents of 1.980 and 8.000 mg/kg, respectively; This study provides technical support for optimizing the cultivation of selenium-rich Morchella sextelatam and developing selenium-rich products in the future.
