番茄枯萎病拮抗细菌S13产芽孢发酵培养基及发酵条件优化

摘要: [目的]提高番茄枯萎病菌拮抗细菌解淀粉芽孢杆菌(Bacillus amyloliauefaciens)S13的芽孢数量和芽孢产率.[方法]采用单因素和正交试验,研究不同的氮源、碳源、pH、温度、培养时间对解淀粉芽孢杆菌形成芽孢的影响.[结果]确定解淀粉芽孢杆菌产芽孢培养基的最佳组分为(g/L):玉米粉0.4,酵母膏0.1,玉米浆1.25,KH2P040.1,K2 HP04 0.1,MgS04.7H20 0.05.对发酵条件进行了研究,产芽孢最适pH为7.0,最适温度为35℃,最适时间为26 h.[结论]通过优化培养基组分和培养基条件,1 000 L罐发酵液中芽孢数达到4.9×109cfu/mL,芽孢产率为90;,为进一步该菌株在复合生物肥中的应用打下基础.
