摘要: 以药用植物刺山柑初秋茎段为外植体,研究了不同灭菌时间对刺山柑无菌外植体建立的影响及不同激素、不同浓度组合对刺山柑丛生芽的诱导、增殖、壮苗和生根的影响.结果表明:外植体灭菌采用0.1;升汞消毒8 min效果最佳,其污染率和死亡率分别为5.6;和6.3;;诱导丛生芽的最适培养基为MS+6-BA 0.6 mg/L,其侧芽萌发率可达90;;增殖的最佳培养基为MS+6-BA 0.6 mg/L+IAA 0.2 mg/L,其增殖倍数可达到4.5倍;壮苗培养基以MS+NAA 0.2 mg/L+GA3 0.2 mg/L效果较好,长势比较旺盛,平均株高、平均芽数均较高;诱导生根的最佳培养基为MS+IBA 0.5 mg/L+0.1;活性炭,生根率达65;,基部几乎无愈伤,且根数较多.
