Rot pathogenic bacteria of Isatis indigotica Fort.: isolation and identification, biological characteristics

  • Objective To isolate and identify the pathogenic bacteria of the root rot of Isatis indigotica Fort. in Xinjiang.
    Methods The root of Isatis indigotica Fort. were collected from Habahe County and QinHe County, ltacy region, Xinjiang, using tissue isolation and dilution coating isolation methods, and the pathogenic bacteria were identified by pathogenicity determination, morphology, molecular biology of ITS sequences and physiological and biochemical properties.
    Results Six fungi strains were separated from the rot root of Isatis indigotica Fort., the strains B4, B7 and B14 were Fusarium solani, Fusarium solani, which were the dominant pathogen with strong pathogenicity; strain B10 was Fusarium oxysporum, strain B16 was Alternaria alstroemeriae, and strain B17 was Chaetomium globosum. Biological characterization showed that all the carbon and nitrogen sources for the test could be utilized as a single source of carbon and nitrogen by these six strains of bacteria, and they could be grown at temperatures ranging from 25~37 ℃ and pH 3~10.
    Conclusion The main pathogens of the root rot of Isatis indigotica Fort. were Fusarium solani, Fusarium oxysporum in Xinjiang, and Alternaria alstroemeriae and Chaetomium globosum were reported as root rot pathogens for the first time in our country. It provided the research basis for rapid diagnosis and prevention of the root rot of Isatis indigotica Fort. in Xinjiang.
